Srs Microscopy

SRS microscopy, or stimulated Raman scattering microscopy, is a label-free imaging technique that maps molecular composition by detecting vibrational signatures within biological samples. It uses synchronized pump and Stokes laser beams whose frequency difference matches a molecular vibration; this resonance produces stimulated Raman gain or loss, which is measured as a rapid change in the transmitted light intensity. In biology, SRS microscopy enables chemically specific imaging of lipids, proteins, metabolites, and other constituents in living cells and tissues without fluorescent labels. Its speed, molecular contrast, and compatibility with live specimens support studies of cell organization, metabolism, disease processes, and tissue structure.

Correlative Light- and Electron Microscopy Using Quantum Dot Nanoparticles

A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of epoxy embedded human pathology tissue. We employ commercial antibody fragment conjugated QDs that are visualized by widefield fluorescence light microscopy and transmission electron microscopy.

Combinatorial 5 fluorescent proteins marking of hematopoietic stem and progenitor cells allows in vivo clonal tracking via confocal and two-photon microscopy, providing insights into bone marrow hematopoietic architecture during regeneration. This method allows non-invasive fate mapping of spectrally-coded HSPCs-derived cells in intact tissues for extensive periods of time following transplantation.

Microwave Assisted Rapid Diagnosis of Plant Virus Diseases by Transmission Electron Microscopy
