Subretinal Space Access

Subretinal space access is a microsurgical technique used to reach the potential space between the neurosensory retina and retinal pigment epithelium, enabling targeted delivery and manipulation within the eye. Access typically involves creating a controlled retinal opening or bleb, often by introducing a fine cannula through the retina and carefully infusing fluid, cells, or therapeutic agents beneath it. This approach supports research and treatment development for inherited retinal disorders, geographic atrophy, and other conditions requiring localized intervention. By placing materials near photoreceptors and retinal pigment epithelial cells, subretinal delivery can improve tissue targeting while limiting exposure to surrounding ocular structures.

Subretinal injections are the most common technique for delivering large therapeutic agents such as proteins and viral vectors to photoreceptors and the retinal pigment epithelium. An alternative method in mice that successfully targets the subretinal space with minimal collateral damage and fast recovery times is described here.

(2016)This video demonstrates the procedure for delivering a fluorescent dye into the subretinal space of a mouse. The process involves anesthetizing the mouse, preparing the eye, making precise incisions to access the subretinal space, and injecting the dye to create a controlled retinal detachment. This technique allows for targeted...

Here we present a community accepted protocol in multimedia format for subretinally injecting a bolus of RPE cells in rats and mice. This approach can be used for determining rescue potentials, safety profiles, and survival capacities of grafted RPE cells upon implantation in animal models of retinal degeneration.
