Temperature Gradient

A temperature gradient is a measurable change in temperature across space or between defined locations, and it matters because living systems respond to thermal differences. It forms when heat is transferred by conduction, convection, or radiation while unequal heating, cooling, or insulation maintains warmer and cooler regions. In biology, temperature gradients influence enzyme reaction rates, membrane properties, metabolic performance, development, and movement as organisms regulate body temperature or seek favorable conditions. Researchers use controlled gradients to study thermotaxis, species distribution, microbial growth, and physiological stress, linking physical conditions to cellular function and ecological responses.

Temperature Gradient Assay: A Method to Test Temperature Preference in Drosophila Larvae

2023

Drosophila fruit flies can distinguish small changes in temperature and therefore select environments with favorable thermal conditions. This video describes a behavioral assay that tests the temperature preference of Drosophila larvae on a linear thermal gradient.

Gradient Polymerase Chain Reaction to Determine the Optimum Annealing Temperature

2025

This video demonstrates the gradient polymerase chain reaction for optimization of the annealing temperature of primers, which sets gradient annealing temperature along wells. The effect of different annealing temperatures is determined by the yield of amplified products that helps to optimize the reaction condition.

2026

We introduce a temperature gradient block and detail its construction and utilization. The block provides 22 discrete temperatures, linearly distributed between a user-defined range from 0 to 90 °C. Data collected from this setup enables testing and developing new theories on the temperature response of soil organic matter decomposition rates.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...
