Nucleofection Technique

Nucleofection is a specialized transfection technique that uses electrical pulses to introduce DNA, RNA, or other nucleic acids directly into the nucleus of living cells. The pulses temporarily disrupt the plasma and nuclear membranes, allowing genetic material to enter while carefully controlled electrical conditions and cell-specific buffers support recovery and viability. In biology, nucleofection enables efficient gene delivery into primary cells, stem cells, immune cells, and other cell types that are difficult to modify with conventional methods. It supports transient gene expression, genome editing, and functional studies, although optimization is often required to balance delivery efficiency with cell survival.

Neuroblast migration is a crucial step in postnatal neurogenesis. The protocol described here can be used to investigate the role of candidate regulators of neuroblast migration by employing DNA/small hairpin RNA (shRNA) nucleofection and a 3D migration assay with neuroblasts isolated from the rodent postnatal rostral migratory stream.

Here, we provided a method to achieve stable transfection of chicken Eimeria parasites by nucleofecting sporozoites or second-generation merozoites. Genetically modified eimerian parasites expressing heterologous antigenic genes could be used as vaccine delivery vehicles.
