Tissue Culture Plate

A tissue culture plate is a sterile, multiwell laboratory vessel used to grow and maintain cells or small tissue samples under controlled conditions, making it essential for biological research. Each well provides a separate compartment where cells receive nutrient-rich culture medium and, when needed, attach to a treated surface while temperature, pH, and gas levels are regulated in an incubator. Tissue culture plates support experiments in cell biology, microbiology, drug screening, toxicity testing, and tissue engineering by enabling parallel cultures, controlled comparisons, and direct observation of cellular growth, behavior, and responses.

The floor plate is a crucial structure of the developing spinal cord, providing diffusible signals to progenitors and differentiating neurons. We describe a method to produce floor-plate conditioned medium, to apply it to fresh spinal cord tissue, and to assess the consequences on proteins of interest by biochemistry.

Histotypic Tissue Culture

Although two-dimensional tissue culture has been common for some time, cells behave more realistically in a three-dimensional culture, and more closely mimics native tissue. This video introduces histotypic tissue culture, where the growth and propagation of one cell line is done in an engineered three-dimensional matrix to reach high cell density. Here, we show the harvesting of cells from donor tissue, followed by cell culture on an engineered construct.

Whole organs can be cultured ex vivo using specialized bioreactors with the goal of repairing or replacing entire organs. This method uses a donor organ that is stripped of all cells, leaving behind the three-dimensional structure, which is then repopulated with new cells.
