Tissue Dissection

Tissue dissection is the controlled separation and examination of biological tissues to reveal their structure, organization, and relationships within an organism. Using instruments such as scalpels, forceps, and microdissection tools, researchers isolate tissue regions while preserving key anatomical features for observation, sampling, or further analysis. In biology, dissection supports the study of organ anatomy, tissue architecture, developmental changes, and disease-related alterations. Samples may be examined directly, prepared for histology, or used in molecular and cellular assays, linking visible structures with their physiological functions and underlying biological mechanisms.

Xenopus laevis provides an ideal model system for studying cell fate specification and physiological function of individual retinal cells in primary cell culture. Here we present a technique for dissecting retinal tissues and generating primary cell cultures that are imaged for calcium activity and analyzed by in situ hybridization.
