Tissue Pre-incubation

Tissue pre-incubation is the controlled period in which excised tissue is maintained in a suitable physiological medium before experimental treatment or measurement. During this step, the tissue equilibrates with temperature, pH, oxygenation, nutrients, and ionic conditions, while recovery from dissection and removal of residual handling effects can improve experimental consistency. In biology, pre-incubation supports ex vivo studies of tissue metabolism, signaling, transport, drug responses, and enzyme activity. Carefully defined duration and medium composition help preserve tissue viability and establish a stable baseline, allowing researchers to distinguish responses caused by the test condition from changes associated with tissue isolation.

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.

Once removed from the body, neuronal tissue is greatly affected by environmental conditions, leading to eventual degradation of the tissue after 6 - 8 h. Using a unique incubation method, which closely monitors and regulates the extracellular environment of the tissue, tissue viability can be significantly extended for >24 h.

In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA. Simulating Temperature in a Soil Incubation Experiment

Laboratory soil warming experiments usually employ two or more constant temperatures in multiple chambers. By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.
