Transgenic Plant Regeneration

Transgenic plant regeneration is the process of producing whole, fertile plants from genetically modified plant cells or tissues, making it essential for studying gene function and developing improved crops. After transformation introduces foreign DNA, scientists culture cells under controlled conditions with plant growth regulators that stimulate callus formation, organogenesis, or somatic embryogenesis; selectable conditions help identify cells that retain the transgene. Regenerated plants are acclimatized, propagated, and evaluated to confirm stable gene integration and expression. This approach supports functional genomics, crop improvement, stress-tolerance research, and the development of plants with valuable agricultural or pharmaceutical traits.

We present an efficient, optimized protocol for Agrobacterium-mediated transformation of an asexual amphibious Brassicaceae species, Rorippa aquatica, without resource-intensive sterile tissue culture procedures. Utilizing a visual transformation marker and shoot regeneration pipeline successfully generated near clonal transformants, thereby advancing the future studies of plant's adaptation to the underwater environment.
