Camp Distribution

cAMP distribution describes how cyclic adenosine monophosphate, a ubiquitous intracellular second messenger, is produced, transported, and localized within cells to regulate diverse biological responses. Adenylyl cyclases generate cAMP from ATP, while phosphodiesterases degrade it; localized enzyme activity, diffusion, and binding to effectors such as protein kinase A create distinct signaling compartments rather than a uniform cellular signal. In biology, mapping cAMP distribution helps explain how cells coordinate metabolism, gene expression, contraction, secretion, and responses to hormones or neurotransmitters. Studying this spatial organization also supports research into signaling dysfunction and the development of more selective therapeutic strategies.

Due to inherent low signal-to-noise ratio (SNR) of Fӧrster resonance energy transfer (FRET) based sensors, measurement of cAMP signals has been challenging, especially in three spatial dimensions. Here, we describe a hyperspectral FRET imaging and analysis methodology that allows measurement of cAMP distribution in three spatial dimensions.

The protocol presented in this study illustrates the effectiveness of the cAMP Difference Detector In Situ in measuring cAMP through two methods.
