Triton X-100 Incubation

Triton X-100 incubation is a laboratory treatment that uses a nonionic detergent to alter biological membranes, making cellular structures more accessible for analysis. During incubation, Triton X-100 interacts with membrane lipids and partially solubilizes the lipid bilayer, allowing antibodies, stains, or other molecular probes to enter cells and tissues. In biology, this step commonly supports immunofluorescence, immunohistochemistry, and cell-based microscopy by improving intracellular labeling while helping preserve structural features when conditions are carefully controlled. Incubation time, detergent concentration, temperature, and sample composition influence membrane permeabilization and signal quality, making optimization essential for reliable experimental results.

This video demonstrates the immunohistochemical staining for S-100 protein in rectal suction biopsy sections.
