Urethral Orifice

The urethral orifice is the external opening through which the urethra releases urine from the urinary tract, and in males it can also convey semen during ejaculation. Its position and surrounding tissues differ between females and males, while coordinated contraction of the bladder detrusor muscle and relaxation of the internal and external urethral sphincters regulate urine passage through the urethra. Understanding the urethral orifice is central to studying urinary anatomy, continence, sexual physiology, and disorders such as urinary tract infection or obstruction, and it guides clinical examination, catheter placement, and other procedures.

In the present protocol, a urethral stricture induction was developed in Wistar rats, followed by urethral reconstruction with a buccal mucosa graft. A retrograde urethrogram and laser Doppler assessment were performed, validating urethral reconstruction (after stricture formation) and graft placement.

Nerve-sparing Mid-urethral Obstruction (NeMO) in Female Small Rodents

Traditional modeling of partial bladder outlet obstruction in rodents is fraught with animal mortality. A denervation injury from dissection around the proximal urethra and bladder neck is also of major concern. We developed and evaluated a safe and reliable mid-urethral obstruction model, avoiding the shortcomings of the traditional model.

Establishment of a Urinary Tract Infection Model in Female Mice

2025

This video demonstrates the establishment of uropathogenic Escherichia coli infection in a female mouse bladder through transurethral catheter instillation. The injected bacteria successfully establish a urinary tract infection within the bladder, providing a valuable platform for studying healthcare-associated diseases related to the urinary tract.

Delivery of DNA Plasmids into Urothelium of Mouse Bladder: An Electroporation-based Procedure to Efficiently Deliver DNA Plasmids into Urothelial Cells of Murine Bladder to Generate Genetically Engineered Models
