Vibratome Slicer

A vibratome slicer is a precision instrument that produces thin sections of biological tissue for microscopic analysis while minimizing the distortion caused by conventional cutting methods. It works by moving a vibrating blade through a stationary tissue block, often embedded in agarose and kept chilled or immersed in buffer, to generate uniform sections without requiring paraffin embedding or freezing. Vibratome slicing is widely used in histology, neuroscience, developmental biology, and anatomy to preserve tissue architecture, cellular relationships, and delicate structures. The resulting sections support immunohistochemistry, electrophysiology, imaging, and three-dimensional reconstruction of biological specimens.

A simple procedure of vibratome sectioning the organ of Corti, followed by immunohistochemistry and confocal microscopy is described. This procedure allows for improved preservation of the fine cytoarchitecture of the mammalian organ of Corti, and consequently allows for accurate quantification of cell types.

Presented is a protocol for the isolation of human and animal ventricular cardiomyocytes from vibratome-cut myocardial slices. High yields of calcium-tolerant cells (up to 200 cells/mg) can be obtained from small amounts of tissue (<50 mg). The protocol is applicable to myocardium exposed to cold ischemia for up to 36 h.
