Viral Genome Labeling

Viral genome labeling is the process of attaching detectable markers to viral DNA or RNA so researchers can identify, track, and measure viral genetic material. Labels may be introduced during genome synthesis by incorporating modified nucleotides, or added afterward through chemical or enzymatic reactions that selectively target nucleic acids. Fluorescent, radioactive, or affinity-based markers can reveal genome localization, replication, packaging, and delivery into host cells. In biology, these approaches support studies of infection dynamics, viral assembly, genome trafficking, and gene-transfer systems, while helping distinguish viral genomes from host nucleic acids and evaluate the performance of antiviral or delivery strategies.

Source: Dembowski, J. A., Deluca, N. A. Purification of Viral DNA for the Identification of Associated Viral and Cellular Proteins. J. Vis. Exp. (2017)This video demonstrates the infection of stationary-phase mammalian cells with HSV-1 (Herpes Simplex Virus-1), followed by replication of the viral genome using host and viral replication machinery. A synthetic nucleoside analog is incorporated into newly synthesized viral DNA to label replicating genomes, which are subsequently isolated for...

Begin with a BACmid, a bacterial artificial chromosome that can replicate like a plasmid inside bacteria. The BACmid contains the entire genome of the vaccinia virus with targeted modifications, but it is not infectious as it requires viral proteins to function.Take a helper virus, which is an attenuated vaccinia virus encoding the gene for EGFP.Infect epithelial cells with the helper virus and simultaneously transfect with the BACmid. The virus expresses EGFP and provides the viral proteins,...

Size and Structure of Viral Genomes

2025

Viral genomes exhibit remarkable diversity in size, structure, and composition, influencing their replication strategies and interactions with host cells. These genomes consist of either DNA or RNA and may be linear or circular. Additionally, they can be single-stranded or double-stranded, with each configuration affecting how the virus propagates within a host. RNA viruses, for instance, generally have smaller genomes than DNA viruses, a factor that contributes to their high mutation rates and...

Isolation and Genome Analysis of Single Virions using 'Single Virus Genomics'

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.
