Viral Rna Silencing Suppressor

A viral RNA silencing suppressor is a virus-encoded protein that counteracts RNA silencing, a cellular defense mechanism that detects and degrades viral RNA. These suppressors act at different stages of the pathway by binding double-stranded RNA or small interfering RNAs, preventing their processing, sequestration, or incorporation into Argonaute-containing RNA-induced silencing complexes. In plant and animal biology, studying these proteins reveals how viruses evade host immunity and how RNA interference regulates gene expression. Viral RNA silencing suppressors also serve as research tools for dissecting silencing pathways and improving applications such as functional genomics, crop protection, and virus-resistant biotechnology.

(2020)This video demonstrates a co-infiltration assay for identifying pathogen effector proteins that suppress RNA silencing in Nicotiana benthamiana 16c. A fluorescent reporter and a candidate effector gene are delivered into plant leaves via Agrobacterium. Normally, RNA silencing degrades reporter transcripts, reducing fluorescence. Suppressor effectors...

Detection of Viral RNA by Fluorescence in situ Hybridization (FISH)

2012

A fluorescence in situ hybridization (FISH) method was developed to visually detect viral genomic RNA using fluorescence microscopy. A probe is made with specificity to the viral RNA that can then be identified using a combination of hybridization and immunofluorescence techniques. This technique offers the advantage of identifying the localization of the viral RNA or DNA at steady-state, providing information on the control of intracellular virus trafficking events.
