Virgin Adults

Virgin adults are sexually mature individuals who have not engaged in sexual intercourse, although virginity is a social and behavioral descriptor rather than a distinct biological state. During puberty, activation of the hypothalamic-pituitary-gonadal axis stimulates gonadotropin release, sex-hormone production, and gamete maturation, so reproductive function can develop independently of sexual experience. In biology, distinguishing sexual maturity from mating history helps researchers study reproductive physiology, sexual behavior, fertility, and developmental outcomes. The concept is also relevant when selecting experimental groups, interpreting animal-model data, and discussing reproductive health without treating sexual experience as a direct measure of biological capability.

This step-by-step protocol analyzes Drosophila negative geotaxis behavior using an automated multi-cylinder system that hosts hundreds of flies and synchronizes their action by an electric motor. Upon synchronization, fly negative geotaxis behavior is assayed, digitally recorded, and analyzed using the self-designed RflyDetection software.

Dissection of the Adult Zebrafish Kidney

The zebrafish kidney is home to both renal and hematopoietic adult stem/progenitor cells, and represents an outstanding opportunity to study these cell types and their progeny in a vertebrate model organism. Here, we demonstrate a detailed dissection procedure that enables the researcher to identify and surgically remove the adult zebrafish kidney, which can be used for applications such as cell isolation, transplantation, and expression studies of kidney and/or blood cell populations.

Mapping and Application of Enhancer-trap Flippase Expression in Larval and Adult Drosophila CNS

We describe a Flippase-induced intersectional Gal80/Gal4 repression (FINGR) method, allowing tissue-specific FLP to determine Gal80 expression patterns. Wherever Gal4 and FLP overlap, Gal4 expression is turned on (Gal80 flipped out) or off (Gal80 flipped in). The FINGR method is versatile for clonal analysis and neural circuit mapping.
