Virion Labeling

Virion labeling is the attachment of detectable markers to complete virus particles, enabling researchers to identify, track, and quantify virions in biological systems. Labels such as fluorescent probes, antibodies, or chemically modified components can bind capsid or envelope structures, or become incorporated into viral proteins or nucleic acids during particle production. Researchers use labeled virions to visualize cell attachment, entry, intracellular trafficking, and interactions with host tissues through microscopy or other detection methods. These approaches connect particle behavior with infectivity and distribution, supporting studies of viral life cycles, host-pathogen interactions, antiviral strategies, and the development of virus-based delivery systems.

Flow Virometry to Analyze Antigenic Spectra of Virions and Extracellular Vesicles

Viruses or extracellular vesicles were immunocaptured with 15 nm magnetic nanoparticles coupled to antibodies recognizing surface antigens. The captured virions or vesicles were labeled with fluorescent antibodies against other surface antigens. The resultant complexes were separated in high magnetic field and analyzed with conventional flow cytometers triggered on fluorescence.

Isolation and Genome Analysis of Single Virions using 'Single Virus Genomics'

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.
