Virus Generation

Virus generation is the controlled production of viral particles in living host cells, a process that supports research in virology, biotechnology, and medicine. It occurs when a viral genome directs infected cells to replicate its genetic material, synthesize viral proteins, and assemble new particles, which may be released through cell lysis or other release pathways. Laboratory virus generation enables researchers to study viral replication and host–pathogen interactions, evaluate antiviral compounds, and produce viral vectors for gene delivery. Careful control of host cells, culture conditions, and viral components helps yield reproducible preparations for experimental and therapeutic applications.

Begin with a microplate containing host cells susceptible to vesicular stomatitis virus (VSV) infection.Introduce a transfection mixture containing the genome plasmid, helper plasmids, and a transfection reagent that forms complexes with the plasmids. Incubate the cells to facilitate plasmid uptake. The genome plasmid encodes a modified VSV genome in which the VSV glycoprotein (VSV-G) gene is replaced with one encoding an envelope glycoprotein (Env) from another virus. The helper plasmids provide...

For in-depth mechanistic analysis of the respiratory syncytial virus (RSV) RNA synthesis, we report a protocol of utilizing the chaperone phosphoprotein (P) for coexpression of the RNA-free nucleoprotein (N0) for subsequent in vitro assembly of the virus-specific nucleocapsids (NCs).

A Technique for the Generation of Influenza Virus-Like Particles via a Mammalian System

This video demonstrates the generation of influenza virus-like particles (VLPs) in a mammalian system. Upon expressing three eukaryotic expression vectors — encoding different viral structural proteins — in suitable mammalian host cells, the expressed viral proteins self-assemble into VLPs on the host plasma membrane and release via budding from the cell surface.
