X-chip Method

The X-ChIP method, or cross-linking chromatin immunoprecipitation, is a molecular biology technique used to identify DNA regions associated with specific proteins in cells. It preserves protein-DNA interactions by chemically cross-linking chromatin, fragments the chromatin, and uses an antibody to immunoprecipitate complexes containing the protein of interest. After reversing the cross-links and purifying the recovered DNA, researchers analyze target regions by quantitative PCR or genome-wide binding patterns by sequencing. X-ChIP helps characterize transcription-factor occupancy, histone modifications, and regulatory elements, providing evidence for how protein-DNA interactions influence gene expression and chromatin organization.
