Cell Extension Quantification

Cell extension quantification is the measurement of protrusions that project from a cell body, such as filopodia, lamellipodia, and broader membrane extensions. In microscopy images, analysis typically identifies cell boundaries and calculates features including extension number, length, area, width, orientation, and the proportion of the cell perimeter involved; time-lapse imaging can additionally track their formation and retraction. In cancer research, these measurements provide objective indicators of cytoskeletal remodeling and cell motility, helping researchers compare migration and invasion across tumor cell types or experimental conditions. The approach can also evaluate how genetic changes, extracellular matrix properties, or candidate drugs alter behaviors associated with metastatic spread.

Using a Macrophage Conditioned Medium to Promote Neuron Extensions

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.

Proboscis extension response or PER is a taste behavior assay that has been used in flies as well as in honeybees. When the proboscis makes contact with an attractive substance, the fly extends its proboscis to consume the substance. Solutions of various sugars are very attractive to the fly.

Primer Extension Capture: Targeted Sequence Retrieval from Heavily Degraded DNA Sources

We present a method of targeted ancient DNA sequence retrieval, which we used to reconstruct the complete mitochondrial genomes of five Neandertal individuals. Comparison of these sequences with present day humans suggests that Neandertals had a long term low effective population size.
