Drg Dissection

DRG dissection is the surgical isolation of dorsal root ganglia, clusters of sensory neuron cell bodies that connect peripheral tissues with the spinal cord. The procedure typically involves exposing the spinal column, removing vertebral tissue, locating the ganglia beside the spinal cord, and carefully separating them for culture, imaging, electrophysiology, or molecular analysis. In cancer research, dissected DRGs provide an experimental system for examining how tumors, metastatic growth, or anticancer treatments influence sensory neurons and contribute to cancer-associated pain. This approach helps link peripheral neural signaling with tumor biology and supports evaluation of potential pain mechanisms and therapeutic interventions.

This video demonstrates a procedure for isolating dorsal root ganglion (DRG) neurons from rat embryos and co-culturing them with Schwann cell precursors to generate mature Schwann cells. The isolated DRGs are processed to isolate neurons and glial cells. The isolated cells are cultured to purify the neurons, which are then co-cultured with Schwann cell-like cells (SCLCs) in a co-culture medium. Interaction with the DRG neurons leads to the maturation of SCLCs into Schwann cells.
