Fluorescent Cre-reporter Allele

A fluorescent Cre-reporter allele is a genetically engineered DNA locus that produces a fluorescent protein in cells where Cre recombinase has acted, enabling permanent visualization of selected cell populations. Typically, Cre excises a loxP-flanked transcriptional stop cassette, placing the reporter under the control of an active promoter; the recombined state is inherited by daughter cells even after Cre is no longer present. In cancer research, this system supports lineage tracing, identification of tumor-initiating or genetically targeted cells, and analysis of clonal expansion, invasion, and metastasis in tissues. Fluorescence-based readouts can connect cell history with tumor development and treatment response.

Genetic associations often remain unexplained at a functional level. This method aims to assess the effect of phenotype-associated genetic markers on gene expression by analyzing cells heterozygous for transcribed SNPs. The technology allows accurate measurement by MALDI-TOF mass spectrometry to quantify allele-specific primer extension products.
