Immunophenotype Gating

Immunophenotype gating is a flow cytometry strategy that identifies and separates cell populations according to their patterns of surface or intracellular markers, making it essential for characterizing heterogeneous samples in cancer research. The process applies sequential gates to measured signals, first excluding debris and nonviable cells, then using marker intensity and combinations to distinguish immune, tumor, or stromal populations. Researchers use immunophenotype gating to quantify cell subsets, assess tumor-associated immune responses, track disease-related changes, and evaluate treatment effects. Careful gate selection and appropriate controls improve the reproducibility and biological interpretation of cellular profiling data.

Whole blood Immunophenotyping is indispensable for monitoring the human immune system. However, the number of reagents required and the instability of specific fluorochromes in premixed cocktails necessitates daily reagent preparation. Here we show that semi-automated preparation of staining antibody cocktail helps establish reliable immunophenotyping by minimizing variability in reagent dispensing.

Ion channels are specialized proteins on the plasma membrane that allow charged ions to pass down their electrochemical gradient. Their main function is to maintain the membrane potential which is critical for cell viability. These channels are either gated or non-gated and can transport more than a thousand ions within milliseconds for the cellular event to occur. Compared to the gated ion channels, the non-gated channels, also known as leakage or passive channels, have no gating mechanism.
