Organoid Histology Processing

Organoid histology processing is the preparation of three-dimensional, cell-derived tissue models for microscopic evaluation of their architecture, cellular composition, and disease features. The workflow typically fixes organoids to preserve structure, embeds them in paraffin or another support medium, sections them into thin slices, and applies stains or antibody-based labeling to visualize nuclei, cell lineages, proliferation, and tumor-associated markers. In cancer research, these methods connect organoid growth in culture with tissue-level evidence, enabling researchers to assess tumor morphology, heterogeneity, invasion-related changes, and responses to drugs or genetic manipulation. Consistent processing also supports comparisons between organoids, patient tumors, and treatment conditions.

Lack of standardization for murine tissue processing reduces the quality of murine histopathological analysis as compared to human specimens. Here, we present a protocol to perform histopathological examination of murine inflamed and uninflamed colonic tissues to show the feasibility of robotic systems routinely used for processing and embedding human samples.

Histological Staining of Neural Tissue

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections. In the study of neuroanatomy, histological techniques are frequently applied to visualize and study nervous system tissue.
