Cholesterol Esters

Cholesterol esters are neutral, highly hydrophobic lipids formed when cholesterol’s hydroxyl group is esterified with a fatty acid, making them important for cholesterol storage and transport. During esterification, a fatty acid is linked to the hydroxyl group through an ester bond, reducing cholesterol’s polarity and allowing the product to accumulate in lipid droplets or occupy the core of lipoprotein particles. In chemistry and biochemistry, cholesterol esters are studied to understand lipid partitioning, membrane and lipoprotein composition, and cholesterol homeostasis; their altered formation or breakdown can influence cellular lipid balance and disease research.

Triglycerides Triglycerides are triester molecules composed of a glycerol backbone that is bound to 3 fatty acids. These form the basis of many biological lipids that are found in vegetable oils and fats. Lipids that are isolated from plant material are the basis of vegetable oils, whereas lipids extracted from animals are used as lard or general sources of fats. A fatty acid molecule has a long carbon chain — between 12 and 20 carbons — with a weak organic acid at one end. Some fatty acids...

Source: Lara Al Hariri and Ahmed Basabrain at the University of Massachusetts Amherst, MA, USA Preparation of SoapExpand Soap is prepared using a saponification reaction, where a base catalyzes the hydrolysis of three ester groups of an oil, such as coconut oil. During saponification, hydroxide ions from the base attack the carbonyl group on the oil to form a ratio of three molecules of soap to one molecule of glycerol. The resulting soap molecule is a long carbon chain, which is...

In this article, a protocol for infection of macrophages with Cryptococcus neoformans is described. Also, a method for sterol depletion from the macrophages is explained. These protocols provide a guide to study fungal infections in vitro and examine the role of sterols in such infections.

Cholesterol: Significance and Regulation

Although not a source of energy, cholesterol plays a significant role as a foundational structure for bile salts, steroid hormones, and vitamin D, as well as being a crucial component of plasma membranes. Approximately 15% of blood cholesterol is derived from our diet, with the remainder synthesized from acetyl CoA by the liver and intestines. Cholesterol is eliminated from the body through its conversion into bile salts, which are eventually discarded in the feces. Considering cholesterol and...
