Molecular Libraries

Molecular libraries are organized collections of distinct molecules assembled to represent chemical diversity and support systematic study of structure and function in chemistry. By varying molecular scaffolds and functional groups through parallel or combinatorial synthesis, researchers generate compounds with different physicochemical properties, then catalog and screen them for binding, reactivity, or other measurable behavior. These libraries accelerate hit identification in drug discovery, help map structure–activity relationships, and provide starting points for optimizing potency, selectivity, and stability. In broader chemical research, they enable reproducible comparisons and data-driven links between molecular structure and observed performance.

Molecular Evolution of the Tre Recombinase

Here we report the generation of Tre recombinase through directed, molecular evolution. Tre recombinase recognizes a pre-defined target sequence within the LTR sequences of the HIV-1 provirus, resulting in the excision and eradication of the provirus from infected human cells. While still in its infancy, directed molecular evolution will allow the creation of custom enzymes that will serve as tools of molecular surgery and molecular medicine.

Primer-Free Aptamer Selection Using A Random DNA Library

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.

Construction of a fosmid library with environmental genomic DNA isolated from the vertical depth continuum of a seasonally hypoxic fjord is described. The resulting clone library is picked into 384-well plates and archived for downstream sequencing and functional screening by the application of an automated colony picking system.
