On-resin Peptide Functionalization

On-resin peptide functionalization is a chemistry technique that modifies a peptide while it remains covalently attached to a polymer support during solid-phase peptide synthesis. After each coupling or selective deprotection step, reagents can react with the peptide’s free N-terminus or exposed side-chain groups, while protecting groups control chemoselectivity and the resin environment supports reagent removal by filtration and washing. This approach enables efficient installation of fluorescent labels, lipid chains, chemical handles, post-translational modifications, and cyclization precursors before final cleavage from the resin. On-resin modification therefore expands access to tailored peptides for biochemical studies, drug discovery, and materials research.

Techniques to dissect the mechanisms underlying the secretion of HIV-1 Nef in exosomes are described. Specific short peptides derived from Nef and protein transfection were exploited to determine the structure, function, and binding partners of Nef’s Secretion Modification Region. These procedures have general relevance in many mechanistic studies.
