Take a liquid impinger device that has been used to capture RNA viruses from bioaerosols.
The impinger liquid contains an anion exchange resin. The resin has positively charged functional groups that concentrate the negatively charged viral particles by electrostatic attraction.
Transfer the liquid to a tube and allow the resin to settle.
Remove the liquid.
Add a virus lysis buffer containing carrier RNA and incubate.
The buffer lyses the viral particles while the carrier RNA acts as a co-precipitant, facilitating recovery of viral RNA during RNA isolation.
Collect the lysate and isolate the RNA.
Perform quantitative reverse transcription polymerase chain reaction or qRT-PCR, where first, RNA is converted into complementary DNA.
Then, using viral gene-specific primers, the cDNA is repeatedly amplified to produce multiple DNA copies for quantification.
A positive amplification curve confirms the presence of the virus in the bioaerosol.
