Schiff Base Formation

Schiff base formation is a condensation reaction in which a primary amine reacts with an aldehyde or ketone to produce an imine containing a carbon–nitrogen double bond. The mechanism begins with nucleophilic attack on the carbonyl carbon, followed by formation of a carbinolamine intermediate, proton transfer, and elimination of water; mild acid catalysis and removal of water can promote the equilibrium toward product formation. In chemistry, Schiff bases provide useful intermediates for organic synthesis and versatile ligands for coordination compounds, while their tunable structures support applications in catalysis, materials chemistry, sensing, and the study of molecular recognition.

Periodic Acid Schiff or PAS Staining: A Method to Assess the Structural Deformities of Glomerulus

2023

In this video, we describe the protocol for performing Periodic Acid Schiff (PAS) stain to assess the structural anomalies in the kidney glomerulus. The PAS staining method enables appropriate evaluation of the glomerulus pathophysiology.

Hydrolysis of a Ni-Schiff-Base Complex Using Conditions Suitable for Retention of Acid-labile Protecting Groups

2017

Here, we present an efficient hydrolysis and subsequent Fmoc protection of an amino acid isolated from a Ni-Schiff-base complex. Hydrolysis conditions presented here are suitable for use when retention of acid-labile side-chain protecting groups is required. This technique may be adaptable to a variety of unnatural amino acid substrates.

Periodic Acid-Schiff Staining of Cells: An In Vitro Technique to Detect Glycogen Levels in Peripheral Blood Mononuclear Cells

Begin with a bacterial suspension in nutrient media.Add the suspension to a multi-well plate and incubate. The nutrients in the media facilitate bacterial growth and attachment. The adhered bacteria secrete extracellular polymeric substances consisting of polysaccharides, proteins, and DNA to form a layer of biofilm.Wash multiple times with buffer to remove unbound bacteria.Invert the plate to ensure the removal of residual liquid from the well and allow the biofilm to dry.Treat with a fixative...
