Acid-fast Culture

Acid-fast culture is a microbiological method used to grow acid-fast bacteria from clinical specimens, supporting the diagnosis of tuberculosis and other mycobacterial infections. In the laboratory, specimens are processed to reduce contamination, concentrated when appropriate, and inoculated into solid media or liquid culture systems that support slow-growing mycobacteria. Their lipid-rich, mycolic acid-containing cell walls retain primary stain after acid-alcohol treatment, helping laboratories recognize colonies or interpret culture signals as acid-fast organisms. Because culture can confirm viable organisms and provide material for species identification and antimicrobial susceptibility testing, it remains important for patient management, infection control, and surveillance of drug-resistant disease.
