Clearance Estimation

Clearance estimation is the quantitative assessment of how efficiently the body removes a drug or other substance from the circulation, usually expressed as a volume of plasma cleared per unit time. In clinical pharmacokinetics, it is inferred from the relationship between drug dose or input rate and systemic exposure, often using clearance = dose/AUC, while renal clearance can be estimated from urine excretion and plasma concentrations. The result helps characterize renal and hepatic elimination, predict drug concentrations over time, and select or adjust dosing regimens. Accurate estimates support therapeutic drug monitoring and reduce the risk of treatment failure or toxicity.
