Cytotoxic Reactions

Cytotoxic reactions are immune responses that damage or destroy target cells, making them important in host defense, transfusion medicine, and immune-mediated disease. They occur when antibodies or cytotoxic lymphocytes recognize antigens on cell surfaces; antibodies can activate complement or recruit natural killer cells for antibody-dependent cellular cytotoxicity, while cytotoxic T cells induce apoptosis through perforin and granzymes. In clinical settings, these mechanisms help eliminate infected or abnormal cells but can also cause tissue injury, as seen in incompatible blood transfusions, autoimmune hemolytic anemia, and some drug-induced cytopenias. Understanding cytotoxic reactions supports diagnosis, risk assessment, and development of targeted therapies.

An Assay to Evaluate Immunodominance in Cytotoxic T Lymphocyte Responses

2025

The video showcases a flow cytometry-based assay for assaying immunodominance in cytotoxic T lymphocytes or CTL responses. Injected fluorescent target cells exhibit varying fluorescence corresponding to immunogenic epitopes. Post-spleen collection and sorting reveal a reduction in high-intensity fluorescent cells, confirming immunodominance in CTL responses.
