Spatula Brush

A spatula brush is a clinical sampling device used to collect exfoliated epithelial cells from the cervix for cytological evaluation. Its spatula gathers cells from the ectocervical surface, while the brush reaches the endocervical canal and gently rotates to dislodge additional cells; the collected material is then transferred to a glass slide or liquid-based collection medium. This combined approach helps obtain a more representative cervical sample than either component alone. Spatula-brush sampling supports screening for cellular abnormalities, including precancerous changes, and provides foundational material for laboratory assessment in gynecologic and clinical research.

Poly(ethylene glycol) (PEG) brush-arm star polymers (BASPs) with narrow mass distributions and tunable nanoscopic sizes are synthesized in via ring opening metathesis polymerization (ROMP) of a PEG-norbornene macromonomer followed by transfer of portions of the resulting living brush initiator to vials containing varied amounts of a rigid, photo-cleavable bis-norbornene crosslinker.

Here, we present the Spatula Montevideo protocol for vitrifying mouse and sheep pre-implantation embryos, consisting of a homemade spatula and utilizing homemade solutions. Embryos are rapidly equilibrated in two cryoprotectant solutions of increasing concentration, loaded onto the spatula, immersed in liquid nitrogen, and finally sealed with a 0.5 mL straw.

Brush cells are rare cholinergic chemosensory epithelial cells found in the naïve mouse trachea. Due to their limited numbers, ex vivo evaluation of their functional role in airway immunity and remodeling is challenging. We describe a method for isolation of tracheal brush cells by flow cytometry.
