Cd31 Positive Cell Isolation

CD31-positive cell isolation is a technique that enriches cells expressing CD31, a surface marker commonly used to identify vascular and endothelial populations for developmental biology research. The method uses antibodies that recognize CD31 on the cell membrane, allowing labeled cells to be selectively retained or sorted from a mixed suspension through an antibody-based separation process. Researchers can then examine enriched populations to study endothelial specification, vessel formation, tissue organization, and changes in cell composition during development. By improving access to relatively defined cell populations, this approach supports downstream culture, molecular profiling, imaging, and functional analyses of developmental processes.

Purification of CD31+ Endothelial Precursor Cells Using Magnetic-Activated Cell Sorting

2025

This video demonstrates a technique for purifying CD31+ endothelial progenitor cells (EPCs) from a culture using magnetic-activated cell sorting (MACS). The cell suspension is treated with a fluorophore-conjugated antibody that binds to CD31 on the EPCs. Completing the labeling of CD31+ EPCs involves adding an antibody complex, which binds to the fluorophores, and a polymer-coated magnetic nanoparticle, which binds to the antibody complex. The labeled cells are then purified using a magnetic...

The differentiation of white and beige adipocytes from adipose tissue vascular progenitors bears potential for metabolic improvement in obesity. We describe protocols for a CD34+CD31+ endothelial cell isolation from human fat and for a subsequent in vitro expansion and differentiation into white and beige adipocytes. Several downstream applications are discussed.
