Collagen Secretion

Collagen secretion is the cellular process that produces and exports collagen, the primary structural protein of the extracellular matrix, enabling tissues to acquire strength, shape, and mechanical support. Cells synthesize procollagen in the endoplasmic reticulum, where selected amino acids undergo hydroxylation, then modify and package it through the Golgi apparatus for secretion by exocytosis. Outside the cell, enzymes remove terminal propeptides, allowing collagen molecules to assemble into fibrils and larger fibers. In developmental biology, this process coordinates extracellular matrix formation during tissue morphogenesis, cell migration, and organ development, while disruptions can alter tissue architecture and developmental outcomes.

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.
