Fibroblast Differentiation

Fibroblast differentiation is the developmental process by which progenitor cells acquire the identity and functions of fibroblasts, connective-tissue cells that build and organize the extracellular matrix. In response to local signals such as transforming growth factor beta and changes in tissue environment, mesenchymal cells activate fibroblast-associated gene programs and begin producing structural proteins including collagen and fibronectin. In developmental biology, studying this process clarifies how connective tissues form, mature, and repair after injury. It also supports research into wound healing, tissue engineering, organ development, and pathological fibrosis, where abnormal fibroblast activation can disrupt normal tissue architecture.

2011

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Generating Neurons by Reprogramming Human Dermal Fibroblast Cells

2025

The video demonstrates the generation of neurons from adult human dermal fibroblasts (aHDFs). The aHDFs are reprogrammed using a lentiviral vector carrying genes for neuronal transcription factors and short hairpin RNAs (shRNAs). The shRNAs inhibit the expression of a neuronal transcriptional repressor, and the neuronal transcription factors activate genes to differentiate aHDFs into neurons. These reprogrammed aHDFs are cultured in an early and a late neuronal conversion medium to generate...

Generating a 3D Co-Culture Model of Human Corneal Fibroblasts and Neurons

2025

This video demonstrates a technique to generate a three-dimensional co-culture model system of primary human corneal fibroblasts and differentiated neuronal cells. This co-culture system can be used to study the stromal-nerve interactions.

Rapid Culturing of Primary Murine Melanocytes and Fibroblasts: Isolating Melanocytes and Fibroblasts from Whole Murine Skin Sample

2023

This video describes the rapid method to isolate and culture the melanocytes and fibroblast simultaneously from the postnatal mouse pup’s skin. These cells can be maintained and manipulated in vitro to study skin cell biology, wound healing, and melanoma.
