Fluorescent Marker Strains

Fluorescent marker strains are genetically engineered organisms that produce fluorescent proteins in selected cells or tissues, making developmental events visible in living specimens. In developmental biology, regulatory DNA sequences can place fluorescent protein genes under the control of tissue-specific or stage-specific promoters; excitation light then elicits emission that reports gene activity, cell identity, position, or lineage over time. These strains support real-time imaging of embryogenesis, tissue formation, cell migration, and morphogenesis, while enabling comparisons between normal and perturbed development. Because fluorescence can be monitored repeatedly, marker strains connect molecular regulation with changing anatomy and help reveal how cells organize into functional tissues.

A procedure for liquid-based culturing and dispensing of C. elegans strains expressing fluorescent reporter proteins is described that does not require expensive sorting equipment. This approach can be applied to numerous inducible C. elegans genes for drug discovery or biosensing of contaminants.

Monitoring Intraspecies Competition in a Bacterial Cell Population by Cocultivation of Fluorescently Labelled Strains

Bacteria may accumulate either detrimental or beneficial mutations during their lifetime. In a population of cells individuals that have accumulated beneficial mutations may rapidly outcompete their fellows. Here we present a simple procedure to visualize intraspecies competition in a bacterial cell population over time using fluorescently labeled...
