Hepatocyte Ablation

Hepatocyte ablation is the targeted removal or destruction of liver parenchymal cells, a method used to examine how tissues respond to cell loss and restore function. It typically relies on genetic, chemical, or physical approaches that selectively injure hepatocytes, triggering coordinated signaling, inflammation, and compensatory proliferation in surviving cells. In developmental biology, this model helps researchers investigate liver formation, cellular plasticity, regeneration, and interactions between hepatocytes and the surrounding tissue. By controlling the timing and extent of ablation, studies can reveal how developmental programs are reactivated after injury and identify mechanisms that maintain liver structure and function.

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.

Sustained fibrosis with deposition of excessive extracellular matrix proteins leads to cirrhosis. Alcohol abuse is one of the main causes of severe liver disease. We established an ethanol-induced zebrafish fibrotic liver model to study the mechanisms and strategies of promoting hepatocyte regeneration upon alcohol-induced injury.

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.
