Upright Droplet Technique

The Upright Droplet Technique is a microscale method for positioning and maintaining small biological samples in an optically accessible droplet, enabling controlled observation during development. A defined volume of culture medium is placed on a support so surface tension preserves the droplet’s shape and keeps embryos or cells localized for imaging, manipulation, or treatment. In developmental biology, this approach can simplify sample handling while limiting reagent use and supporting continuous visualization of growth, morphogenesis, and cell behavior. Its compact format is useful for experiments that require precise environmental control, repeated measurements, or parallel testing of developmental conditions.

The ex vivo upright droplet culture is an alternative to current in vitro and in vivo experimental techniques. This protocol is easy to perform and requires smaller amounts of reagent, while permitting the ability to manipulate and study fetal vascularization, morphogenesis, and organogenesis.

Two techniques for isolating cellular lipid droplets from 1) yeast cells and 2) human placentas are presented. The centerpiece of both procedures is density gradient centrifugation, where the resulting floating layer containing the droplets can be readily visualized by eye, extracted, and quantified by Western Blot analysis for purity.
