In Utero Study

An in utero study examines embryonic or fetal development within the uterus, preserving the maternal, placental, and physiological context that shapes development. Researchers use approaches such as prenatal imaging, tissue sampling, or controlled manipulation to monitor developmental processes and assess how signals, genes, nutrients, or environmental exposures influence growing organs and tissues. In developmental biology, these studies reveal interactions between the fetus, placenta, and maternal environment that may be missed in isolated cell or ex utero models. They support investigation of congenital disorders, developmental timing, prenatal therapies, and the effects of maternal health or environmental conditions on offspring outcomes.

This manuscript provides protocols that use in utero electroporation (IUE) to describe the structural connectivity of neurons at the single-cell level and the excitability of fluorescently labeled neurons. Histology is used to characterize dendritic and axonal projections. Whole-cell recording in acute slices is used to investigate excitability.

Utero-tubal embryo transfer uses the utero-tubal junction as a barrier to prevent the embryo outflow that may occur when performing uterine transfer. Vasectomized males are required to obtain pseudopregnant recipients for embryo transfer. Both techniques are discussed.
