Intra-amniotic Injection

Intra-amniotic injection is a technique that delivers a defined substance into the amniotic cavity surrounding a developing embryo or fetus, enabling controlled study of prenatal exposure and development. Using a fine needle, researchers introduce a measured volume into the amniotic fluid, where the injected material can contact embryonic tissues, be swallowed, or distribute through the surrounding fluid. In developmental biology, this approach helps assess how drugs, nutrients, signaling molecules, or potentially harmful compounds influence organ formation, growth, and tissue patterning. It also supports experimental models of developmental toxicity and provides a controlled way to investigate mechanisms that shape embryonic development.

Encephalopathy of prematurity encompasses the central nervous system abnormalities associated with injury from preterm birth. This report describes a clinically relevant rat model of in utero transient systemic hypoxia-ischemia and intra-amniotic lipopolysaccharide administration (LPS) that mimics chorioamnionitis, and the related impact of infectious stimuli and placental underperfusion on CNS development.

We describe a protocol for performing an in utero transplantation (IUT) through intravenous and intra-amniotic routes of injection in the murine model. This protocol can be used to introduce cells, viral vectors, and other substances into the unique immune-tolerant fetal environment.

Lymph nodes are the immunological tissues that orchestrate immune response and are a critical target for vaccines. Biomaterials have been employed to better target lymph nodes and to control delivery of antigens or adjuvants.
