Nocodazole Washout

Nocodazole washout is a laboratory procedure that removes the microtubule-disrupting drug nocodazole from cultured cells to study the timing and consequences of microtubule recovery. Nocodazole promotes microtubule depolymerization, and replacing the drug-containing medium with fresh medium allows microtubules to regrow and reorganize under controlled conditions. In developmental biology, this approach helps researchers examine cell-cycle progression, mitotic spindle reassembly, chromosome segregation, cell polarity, and changes in cell shape or movement. Comparing cells before and after washout can reveal how cytoskeletal dynamics coordinate tissue formation and developmental transitions, while providing a synchronized experimental window for imaging and molecular analysis.

The first protocol outlines an optimized method for immunolabeling stable and dynamic microtubules. The second protocol provides a method to image and quantify nascent microtubules specifically.

Preparation of Onion Root Tips and Solutions ExpandFor the cell cycle experiment using root tips, first, leave an onion suspended over a beaker of water to grow roots for several days. Then, clean the onion roots of any dirt or debris. Next, in a 1.5 mL tube, dissolve 10 mcg of nocodazole per 1 mL of dimethyl sulfoxide solution, making 1 tube per student. Finally, set out all of the other tools, materials, and reagents from the list of materials onto each student's lab bench.

Studying Cell Cycle-regulated Gene Expression by Two Complementary Cell Synchronization Protocols

We report two cell synchronization protocols that provide a context for studying events related to specific phases of the cell cycle. We show that this approach is useful for analyzing the regulation of specific genes in an unperturbed cell cycle or upon exposure to agents affecting the cell cycle.

We describe a simple approach for diagnosis of halitosis in dogs as well as a dietary approach for its management. This protocol may be extended to the management of halitosis in humans in the near future.
