Zebrafish Mutant Penetrance

Zebrafish mutant penetrance is the proportion of genetically similar mutant fish that display a particular phenotype, making it a key measure of how reliably a developmental mutation produces an observable trait. It reflects interactions between the altered gene and other influences, including genetic background, modifier genes, environmental conditions, and developmental timing, so the same mutation may yield variable outcomes. Measuring penetrance across embryos, larvae, or adults helps researchers distinguish consistent gene functions from variable effects, interpret genotype–phenotype relationships, and assess the robustness of zebrafish models for studying organ formation, disease mechanisms, and developmental pathways.

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.

Generation of Maternal Mutants Using zpc:cas9 Knock-in Zebrafish

Here, we describe a protocol for generating maternal mutant that couples a stable zpc:cas9 knock-in line with Tol2-mediated delivery of sgRNA expression cassettes.

Lens Transplantation in Zebrafish and its Application in the Analysis of Eye Mutants

Lens development involves interactions with other tissues. Several zebrafish eye mutants are characterized by an abnormally small lens size. Here we demonstrate a lens transplantation experiment to determine whether this phenotype is due to intrinsic causes or defective interactions with tissues that surround the lens.

This protocol describes the synthesis of cyclic cell-penetrating peptides with aromatic cross-links and the evaluation of their permeability across biological barriers.

Protein Phase Separation Assay: An Optogenetic Method for Mutant RNA-Binding Protein Phase Separation in Spinal Motor Neurons of Zebrafish Larvae

In this video, we describe a phase separation assay, wherein intracellular proteins comprising intrinsically disordered regions fused to a photosensitive oligomerization domain are induced via blue-light exposure to self-associate into membrane-less liquid-like condensates.
