Decellularized Matrix

Decellularized matrix is a biological scaffold produced by removing cells and cellular components from tissue while retaining much of its native extracellular matrix, making it valuable for tissue engineering and regenerative medicine. Chemical agents, enzymes, detergents, and physical treatments disrupt cell membranes and degrade nucleic acids, while processing conditions are controlled to preserve structural proteins, biochemical signals, and tissue-specific architecture. The resulting material can support cell attachment, migration, and differentiation and may be recellularized with a patient’s cells to engineer replacement tissues. Its composition and mechanical properties also provide a framework for designing disease models, drug-testing platforms, and biomimetic constructs.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
