Hydrogel Sample Preparation

Hydrogel sample preparation is the controlled process of forming, shaping, conditioning, and handling water-rich polymer networks for engineering experiments and device development. It typically involves dissolving or dispersing polymer precursors, initiating physical or chemical crosslinking, and controlling factors such as composition, temperature, pH, and curing time to obtain samples with reproducible swelling, stiffness, and porosity. Prepared hydrogels can then be cut, molded, hydrated, sterilized, or functionalized for mechanical testing, transport studies, sensing, tissue interfaces, and drug-delivery research. Consistent preparation reduces variability and helps link processing conditions to performance in soft materials and bioengineering applications.

Our laboratory has developed DNA-crosslinked polyacrylamide hydrogels, a dynamic hydrogel system, to better understand the effects of modulating tissue stiffness on cell function. Here, we provide schematics, descriptions, and protocols to prepare these hydrogels.

We present a new polyacrylamide hydrogel, called hydroxy-PAAm, that allows a direct binding of ECM proteins with minimal cost or expertise. The combination of hydroxy-PAAm hydrogels with microcontact printing facilitates independent control of many cues of the natural cell microenvironment for studying cellular mechanostransduction.
