Methane Oxygen Gradient

A methane-oxygen gradient is a spatial change in methane and oxygen concentrations, commonly formed across sediments, soils, water columns, or engineered interfaces, that controls where these gases meet and react. Oxygen typically diffuses from the surface while methane rises from anoxic layers, creating an interface where aerobic methanotrophs oxidize methane to carbon dioxide; below the oxygen limit, anaerobic methane-oxidation pathways may operate when suitable electron acceptors are available. Mapping this gradient helps researchers locate methane-consuming microbial communities, quantify controls on greenhouse-gas transport, and assess emissions from wetlands, lakes, marine sediments, and other environments.

Agarose-Based Model Ecosystem for Cultivating Methanotrophs in a Methane-Oxygen Counter Gradient

A protocol is described for preparing a simple model ecosystem that recreates the methane-oxygen counter gradient found in the natural habitat of aerobic methane-oxidizing bacteria, enabling the study of their physiology in a spatially resolved context. Modifications to common biochemical assays for use with the agarose-based model ecosystem are also described.

The control of chemical and oxygen gradients is essential for cell cultures. This paper reports a polydimethylsiloxane-polycarbonate (PDMS-PC) microfluidic device capable of reliably generating combinations of chemical and oxygen gradients for cell migration studies, which can be practically utilized in biological labs without sophisticated instrumentation.

Characterization of Sickling During Controlled Automated Deoxygenation with Oxygen Gradient Ektacytometry

Here, we present oxygen gradient ektacytometry, a rapid and reproducible method to measure red blood cell deformability in samples from patients with sickle cell disease under controlled deoxygenation and reoxygenation. This technique provides a way to study red blood cell sickling and to monitor sickle cell disease treatment efficacy.
