Non Targeted Analysis

Non-targeted analysis is an analytical strategy that broadly profiles chemical constituents in a sample without limiting measurement to a predefined list of compounds. In environmental research, samples are commonly separated by chromatography and analyzed with high-resolution mass spectrometry, which records signals across a wide mass range; computational workflows then detect, group, and tentatively identify molecular features using accurate mass, isotopic patterns, and fragmentation data. This approach can reveal expected and previously unrecognized contaminants, transformation products, and natural chemicals in water, soil, sediment, and biota. It supports environmental monitoring, pollution-source assessment, exposure evaluation, and prioritization of compounds for confirmatory testing.

Targeted DNA Methylation Analysis by Next-generation Sequencing

Bisulfite amplicon sequencing (BSAS) is a method for quantifying cytosine methylation in targeted genomic regions of interest. This method uses bisulfite conversion paired with PCR amplification of target regions prior to next-generation sequencing to produce absolute quantitation of DNA methylation at a base-specific level.

Here we are presenting a chromatin immunoprecipitation (ChIP) procedure for genome-wide location analysis of protein isoforms that differ in a histone-binding domain. We are applying it to ChIP-Seq analysis to identify the targets of the KDM5A/JARID1A/RBP2 histone demethylase.

Depletion of Mouse Cells from Human Tumor Xenografts Significantly Improves Downstream Analysis of Target Cells

Human tumor xenografts are vascularized and infiltrated by cells of mouse origin during the growth phase in vivo. To circumvent the bias caused by these contaminating cells during downstream analysis, we developed a method allowing for comprehensive depletion of all mouse cells by magnetic cell sorting.
