D-loop Extension

D-loop extension is a DNA synthesis process that lengthens a displacement loop, a transient three-stranded structure formed when one single DNA strand pairs with a complementary sequence in a duplex. After strand invasion creates the D-loop, DNA polymerase uses the exposed complementary strand as a template to extend the invading strand, while the displaced strand remains temporarily unpaired. This mechanism supports homologous recombination and DNA repair by copying genetic information from an intact template, and it also contributes to the study of genome stability, replication intermediates, and programmed genetic exchange. Understanding D-loop extension helps clarify how cells preserve sequence accuracy and respond to DNA damage.

Using a Macrophage Conditioned Medium to Promote Neuron Extensions

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.
