Dna Hybridization Capture

DNA hybridization capture is a molecular technique that selectively isolates genomic regions of interest, enabling targeted analysis within genetics. The method uses labeled or immobilized single-stranded probes with sequences complementary to target DNA; after the sample is denatured, complementary strands anneal under controlled conditions, and probe-bound fragments are recovered while nonspecific DNA is removed through washing. Captured DNA can be amplified and analyzed by sequencing, genotyping, or mutation detection. By enriching specific loci before analysis, the technique reduces sequencing effort, improves sensitivity for rare variants, and supports studies of disease-associated genes, structural variation, and genome organization.

Described is a two-step labeling process using β-glucosyltransferase (β-GT) to transfer an azide-glucose to 5-hmC, followed by click chemistry to transfer a biotin linker for easy and density-independent enrichment. This efficient and specific labeling method enables enrichment of 5-hmC with extremely low background and high-throughput epigenomic mapping via next-generation sequencing.

Here, we present a simple method for performing fluorescence DNA in situ hybridization (DNA ISH) to visualize repetitive heterochromatic sequences on slide-mounted chromosomes. The method requires minimal reagents and it is versatile for use with short or long probes, different tissues, and detection with fluorescence or non-fluorescence-based signals.
