Embryo Electroporation

Embryo electroporation is a gene-delivery technique that introduces DNA, RNA, or genome-editing reagents into early embryos to modify gene activity or inheritance. Brief electrical pulses transiently disrupt cell membranes, creating pores through which charged nucleic acids and associated molecular components enter embryonic cells; treated embryos can then develop while carrying the introduced or edited genetic material. In genetics, the method supports transgenesis, CRISPR-based genome editing, and functional studies of developmental genes in model organisms. By enabling targeted manipulation at early developmental stages, embryo electroporation helps link specific genetic changes to phenotypes and accelerates the generation of research models.

Gene Transfer into Older Chicken Embryos by ex ovo Electroporation

A method of gene transfer into chicken embryos at later incubation stages (older than Hamburger and Hamilton stage (HH) 22) is described. This method overcomes disadvantages of in ovo electroporation applied to older chicken embryos and is a useful technique to study gene function and regulation at older developmental stages.

Chick in ovo electroporation is a technique which allows genetic manipulation of the avian embryo. Common applications of this technique include functional analysis of genes and putative enhancer elements. This video demonstrates neural tube electroporation in HH 10 chick embryos. Injection technique and proper egg handling are discussed.

Embryo Microinjection and Electroporation in the Chordate Ciona intestinalis

We present transient transgenesis and gene knockdown in Ciona intestinalis, a chordate sister group to vertebrates, using microinjection and electroporation techniques. Such methods facilitate functional genomics in this simple invertebrate that features rudimentary characteristics of vertebrates, including notochord and head sensory epithelia, and many orthologs of human disease associated genes.
