Emsa Analysis

EMSA analysis, or electrophoretic mobility shift assay, is a biochemical technique used to detect and characterize interactions between nucleic acids and binding proteins. In a typical assay, a labeled DNA or RNA fragment is incubated with a protein, and the resulting complexes migrate more slowly than free nucleic acid during non-denaturing gel electrophoresis, producing a shifted band. Researchers use EMSA to investigate transcription factor binding, identify regulatory DNA elements, compare binding strength or specificity, and assess the effects of mutations or competing sequences. In genetics, the method helps connect sequence variation with changes in gene regulation and supports studies of transcriptional control.

siRNA Transfection and EMSA Analyses on Freshly Isolated Human Villous Cytotrophoblasts

This protocol describes a method for efficiently transfecting siRNA in freshly isolated human villous cytotrophoblasts using microporation and identifying DNA-protein complexes in these cells. Transfected cells can be monitored by Western blot and EMSA analyses during the 4-day culture time.

Here we present a protocol to analyze RNA/protein interactions. The electrophoretic mobility shift assay (EMSA) is based on the differential migration of RNA/protein complexes and free RNA during native gel electrophoresis. By using a radiolabeled RNA probe, RNA/protein complexes can be visualized by autoradiography.

A Neuronal and Astrocyte Co-Culture Assay for High Content Analysis of Neurotoxicity

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.
